In response to the requirements of customers in south China, huide yi will hold the autumn continuous flow technology training course in the biological incubator of shenzhen high-tech zone from September 19 to 20, 2018.
By then, huideyi will also bring Contichrom Discovery chromatography system, QuikSep EC manual chromatography column, QuikSep SC simple chromatography system and other sets of equipment to appear together, which can provide operation demonstration for the majority of customers.
? ?傳統(tǒng)層析工藝，由于流穿效應(yīng)，有40-60%的填料是無(wú)法被利用的，從而使得載量和處理量大為受限。Capture SMB用于優(yōu)化捕獲環(huán)節(jié)，作為最簡(jiǎn)單和最優(yōu)化的2柱位設(shè)計(jì)，可使純化效率顯著提高，并且使填料用量大為縮減。
? ? ? Capture SMB是將第一根層析柱的抗體/蛋白流穿繼續(xù)上樣至第二根層析柱，待第一根層析柱完全滿載后，方結(jié)束其上樣進(jìn)行淋洗，并繼續(xù)進(jìn)行洗脫、再生、再平衡操作，而此時(shí)第二根層析柱會(huì)獨(dú)立進(jìn)行上樣操作。通過(guò)切換柱位操作，保證2根層析柱的洗脫、再生、上樣等操作交替銜接進(jìn)行，從而實(shí)現(xiàn)工藝與系統(tǒng)硬件設(shè)計(jì)的完美結(jié)合，使填料利用率最大化，累積用量大為縮減。

Mabselect SuRe：靜態(tài)載量60 g/L，使用壽命200 cycles；
處理量：600 kg mAb，填料體積：均為100 L。
MCSGP（多柱逆流溶劑梯度純化）
? ? ??MCSGP（Multicolumn Countercurrent Solvent GradientPurification）用于精純環(huán)節(jié)，可有效克服SMB（模擬移動(dòng)床）僅適用于二元分離、無(wú)法實(shí)現(xiàn)線性梯度的缺點(diǎn)。
? ? ? ?傳統(tǒng)的層析精純工藝，必須在收率和純度之間做出權(quán)衡，兩者此消彼長(zhǎng)。如何在保證獲得高純度產(chǎn)品的同時(shí)，得到高收率，一直是層析精純工藝所面臨的難點(diǎn)。
? ??? MCSGP工藝，可將含有不純組分的產(chǎn)品回收在一個(gè)封閉的環(huán)內(nèi)，進(jìn)行循環(huán)式的層析操作，從而最大限度地提取純產(chǎn)品，使得收率和純度二者兼得。

? ??? ?N-Rich工藝是微量物質(zhì)的富集過(guò)程，比如將產(chǎn)品的相關(guān)雜質(zhì)選擇性地富集在一個(gè)循環(huán)的層析過(guò)程中，待富集至目標(biāo)量，再進(jìn)行洗脫回收。該方法特別適用于分離與產(chǎn)品有關(guān)的雜質(zhì)，如異構(gòu)體和聚集體，并同時(shí)用于發(fā)現(xiàn)研究。

Discovery技術(shù)簡(jiǎn)介
? ? ? Discovery層析技術(shù)可將多步層析整合為一步，從而縮短純化時(shí)間，提高純化效率。第1步層析的產(chǎn)物無(wú)需進(jìn)行收集，通過(guò)在線稀釋，可直接上樣至第2根層析柱。即第1步層析的洗脫與第2步層析的上樣同時(shí)進(jìn)行，使純化周期顯著縮短。ContichromDiscovery系統(tǒng)可額外擴(kuò)展出35個(gè)入口，實(shí)現(xiàn)高通量純化。
應(yīng)用案例
? ? ? 第一步Protein A；第二步CIEX；第三步Desalting（分子篩脫鹽換液）。
? ? ? ?在該過(guò)程中，Protein A的洗脫組分經(jīng)過(guò)在線稀釋，直接上樣至CIEX層析柱，CIEX進(jìn)行線性洗脫，去除降解物及聚集體，將目標(biāo)組分直接上樣至脫鹽柱。三步層析緊密銜接，前一步層析的洗脫與后一步層析的上樣同時(shí)進(jìn)行，使得純化效率顯著提高。
? ? ? ?最終產(chǎn)品經(jīng)SE-HPLC檢測(cè)，純度高達(dá)99.5%，二聚體含量?jī)H為0.5%，比Protein A - Desalting兩步層析所得產(chǎn)品的二聚體含量降低近6倍，該產(chǎn)品可直接用于氨基酸序列測(cè)定、二硫鍵測(cè)定以及糖型分析等高純度要求的檢測(cè)。
? ? ? ?Discovery高通量（HT）型系統(tǒng)，采用Protein A - CIEX- Desalting三步向?qū)K，每次上樣10 mL單抗上清液（1.0 mg/mL），每次產(chǎn)量均≥6.5 mg。單個(gè)樣品的三步純化耗時(shí)約為74 min，在24 h內(nèi)純化出多達(dá)19個(gè)樣品，而且該時(shí)間包含了層析柱的清洗環(huán)節(jié)。該系統(tǒng)持續(xù)運(yùn)行5天，純化樣品總數(shù)量高達(dá)91個(gè)。
